Eugenol targeting CrtM inhibits the biosynthesis of staphyloxanthin in <em>Staphylococcus aureus</em>

Staphylococcus aureus is a serious foodborne pathogen threatening food safety and public health. Especially the emergence of methicillin-resistant aureus (MRSA) increased difficulty S. aureus treatment. Staphyloxanthin is crucial virulence factor aureus. Blocking staphyloxanthin production could help host immune system counteract invading aureus cells. In this study, we first screened for inhibitors using virtual screening method. The outcome method resulted in identification eugenol (300 μg/mL), which significantly inhibits aureus ATCC 29213, aureus Newman, MRSA ATCC 43300 and MRSA ATCC BAA1717 by 84.2%, 63.5%, 68.1%, 79.5%, respectively. growth curve assay, field-emission scanning electron, confocal laser microscopy analyses confirmed that at test concentration did not affect morphology Moreover, survival rate 29213 43300 under H2O2 pressure decreased to 51.9% 45.5% presence eugenol, quantitative RT-PCR molecular simulation studies revealed targets biosynthesis downregulating transcription crtM gene inhibiting activity CrtM enzyme. Taken together, determined was prominent compound inhibitor combat aureus especially infections.
